A seven-month-old baby named Tristan Durtschi hasn't been able to sit up and hold his head up, begins biting his lip and tongue, banging his head, and hitting his face from having a rare disorder called Lesch Nyhan Syndrome. Twenty-six-year-old Janelle King has been dealing with fatigue, tingling that spreads from her toes to the rest of her lower body, back pain, and eventually paralysis of her entire body that restricts her lungs due to a rare inflammatory disorder called Devic's Disease (NMO).

Every year, millions of Americans fall prey to real-life medical mysteries ailments that go undiagnosed or misdiagnosed for years. Their lives are thrown into turmoil as medical professionals struggle to understand their baffling conditions. Mystery diagnosis tells their stories.
